In their first year of college, students Farhan Qureshi and Raju Rastogi join the prestigious Imperial College of Engineering (ICE) in Delhi and meet Ranchhoddas Shamaldas Chanchad (aka Rancho), their roommate. Rancho is passionate about experimenting and consequently tops the class. Rancho's carefree approach to education results in him being at odds with the college's director, Dr. Viru Sahastrabuddhe (nicknamed "Virus" by the students), who believes in a strict and competitive education system.
The film begins with wildlife photographer Farhan Qureshi (R. Madhavan) boarding an airplane from Delhi when he receives a phone-call from Chatur Ramalingam (Omi Vaidya), his biggest rival from college, who claims to be knowing the whereabouts of his long-lost collegemate Ranchhoddas Shyamaldas Chanchad a.k.a. Rancho (Aamir Khan). In hopes of meeting Rancho after 10 years, Farhan fakes a heart attack to create an emergency landing of his flight and runs into another former collegemate Raju Rastogi (Sharman Joshi), who forgets to wear his trousers in the hurry of joining him from outside his house.

When a student named Joy Lobo commits suicide after failing to meet a project deadline, Rancho confronts Virus about the extreme pressure placed on ICE students, but is rudely rebuffed. One night, the trio gatecrashes a wedding and meet Virus' youngest daughter, Pia, only later realizing that the wedding is for Pia's older sister, Mona. Although initially upset by Rancho’s behavior, Pia develops a crush on Rancho’s after he pranks her boyfriend Suhas, showing his materialistic side. Fed up with Suhas' materialistic attitude, Pia angrily breaks up with him. An infuriated Virus warns Farhan and Raju about the effects of them being friends with Rancho by pointing out the modest financial situations of their families, scaring Raju into bunking with Chatur "Silencer" Ramalingam, a competitive, arrogant student who believes in learning by rote memorization. To teach Chatur a lesson about memorization, Rancho and Farhan secretly make obscene modifications to the script of Chatur's teacher's day speech. After a disastrous speech before the teachers, Virus, and the Minister of Education, the embarrassed and furious Chatur makes a bet with Rancho to see who will be more successful after ten years.
The two then rush to the Imperial College of Engineering (ICE) to meet Chatur, who reminds them of the bet that he had made with Rancho 10 years ago about who will end up more successful, and boasts to them about being the prosperous vice president of a reputable company in the United States. Frustrated, Farhan and Raju confront Chatur over his mockery when he reveals that he has traced Rancho to Shimla, Himachal Pradesh. The three set out to find Rancho and Farhan reminisces about their college days during the journey.

The night before their final exams, Raju's paralyzed father experiences a heart attack. With Pia's help, Rancho rushes Raju's father to the hospital on Pia's scooter. After realizing that Rancho saved his father's life, Raju reconciles with him. At the end of the year, Rancho comes first in the class, while Farhan and Raju are last and second to last, respectively. During the yearly class photo, Rancho makes a bet with Virus that if either Raju or Farhan gets a job from on-campus interviews, Virus will shave his mustache.
Over a flashback 10 years prior, it is shown that Farhan and Raju joined the prestigious ICE in their first year of college. Farhan came to study at ICE as his conservative father (Parikshit Sahni) forced him to pursue engineering, while Raju's family hopes that he will bring them out of poverty. The two soon befriend their third roommate Rancho, who is passionate about experimenting using different theories and consequently tops the class. He gives unorthodox answers to the teachers about having a rather understandable and enjoyable education system. Rancho's carefree approach to education results in him being at odds with Dr. Viru Sahastrabuddhe (Boman Irani), the director of ICE who is nicknamed "Virus" by the students. Virus believes in a strict and competitive education system and his draconian teaching philosophy contrasts with Rancho's easygoing lifestyle.

One night in their fourth year of college, Rancho tells his friends why they consistently place last: Farhan's passion is photography, not engineering, and Raju lacks self-confidence. After Farhan and Raju promise to confront their problems if Rancho confesses his feelings for Pia, the three of them drunkenly break into Virus' house. Virus spots them during their escape, recognizing Raju, and the next day gives Raju an ultimatum: to incriminate his friends or be expelled himself. Distraught, Raju attempts suicide but survives, leading to Virus revoking the expulsion. Raju recovers, thanks to the support and care of his friends and family. Raju is successful in a job interview, while Farhan convinces his father to let him become a photographer.
When another passionate student named Joy Lobo (Ali Fazal) is denied graduation by Virus on grounds of failure to meet his project deadline, Rancho decides to cheer him up by completing thr project with the help of Farhan and Raju, but the depressed Joy unfortunately dies of suicide by hanging before the trio's implementation. Rancho confronts Virus about the extreme pressure placed on ICE students, but is strongly rebuffed. Virus sends letters to Farhan and Raju's parents expressing dismay about their children's behaviour in college.

Virus, in an attempt to protect his fragile ego, tries to sabotage Raju's chances of success by holding an unfairly difficult exam. With Pia's help, Rancho and Farhan break into Virus' office and steal the exam paper. Upon being given the paper, Raju refuses to cheat and throws it away, but the three of them are caught and expelled. Pia confronts her father over her brother's suicide, which was prompted by similar pressure placed on him by Virus. That night, during a heavy rainstorm, Mona, who is pregnant, goes into labor. Unable to drive to a hospital, Rancho modifies a vacuum cleaner into a ventouse and delivers the baby with the help of Pia, Farhan, and Raju. A grateful Virus acknowledges Rancho by giving him a valuable space pen, which he promised to give only to his most distinguished student, and revokes the trio's expulsions. On graduation day, Rancho suddenly disappears.
After harsh confrontations on Rancho by Farhan and Raju's parents, the hungry trio gatecrash a wedding and meet Virus' younger daughter Pia (Kareena Kapoor), only later realising that the wedding is for Pia's older sister Mona (Mona Singh). An infuriated Virus warns Farhan and Raju about the effects of them being friends with Rancho, pointing out the modest financial situations of their families. While Farhan remains loyal to Rancho, Raju is provoked to move into the room of Chatur, an arrogant, competitive student, who believes in rote memorization. He is nicknamed "Silencer" by the students as he consumes some antacid pills and farts off in a silent and yet lethal way.

10 years later
Determined to teach Chatur a lesson about rote memorization and also show Raju the reality, Rancho and Farhan secretly make obscene modifications (like "chamatkar" (serve) to "balatkar" (screw) and "dhan" (money) to "stan" (bossoms)) to the script of Chatur's Teachers' Day speech dedicated to Virus. After a disastrous speech before the teachers, Virus, and the Minister of Education, the embarrassed and furious Chatur makes the bet with Rancho to see who will end up more successful after 10 years.
Chatur, now the prosperous vice president of a reputable company in California, calls Farhan and Raju to remind them of the bet he made with Rancho about who would be more successful. The three of them set out to find Rancho. Upon reaching Shimla, where Chatur has located Rancho's house, they come across a different man named Ranchhoddas Shyamaldas Chanchad, whose face is pasted over Rancho's in their graduation portrait. The man is reluctant to reveal anything at first and threatens the trio at gunpoint, only relenting after Farhan and Raju threaten to flush his late father's ashes down a toilet. He explains that the "Rancho" at ICE was his family's gardener's son "Chhote", who was incredibly intelligent and gifted. Ranchhoddas' father paid for Chhote to attend ICE, on condition that he use his son's name and, after graduating, cut all contact with anyone at ICE. Chhote, who was more interested in learning than in getting a degree, agreed, but afterward warned that two idiots would come looking for him one day. Ranchoddas gives them Chhote/Rancho's address in Ladakh.

On the way there, Farhan and Raju gatecrash Pia's wedding to her materialistic boyfriend Suhas in Manali and convince her to come with them to find Rancho. At the address in Ladakh, the group is astonished to find a thriving school. They meet "Millimetre", formerly an errand boy at ICE, now Rancho's assistant, who tells them that Rancho has keenly followed their careers. They reunite with Rancho on a sandbar. Rancho admits he is still in love with Pia and the two share a kiss. Chatur assumes that Rancho is a mere schoolteacher and mocks him, but Rancho reveals himself to be Phunsukh Wangdu, a successful inventor with 400 patents, whom Chatur's company is courting. A flabbergasted Chatur accepts defeat as the others run away from him laughing.
Although initially upset by Rancho's behaviour, Pia eventually develops a crush on him after he pranks her shallow, materialistic boyfriend Suhas Tandon (Sanjay Lafont) twice, showing his materialistic side. Firstly, Rancho spills mint chutney on Suhas' expensive shoes at Mona's wedding ceremony earlier. Secondly, he fools Suhas that the expensive watch he had gifted to Pia has gone missing. Fed up with Suhas' materialistic attitude, Pia angrily breaks up with him.

On the same night, Raju's paralyzed father (Mukund Bhatt) suffers from a medical emergency and is rushed to the hospital by Rancho via Pia's scooter. After hearing how the timely intervention saved his father's life, Raju reconciles with Rancho and Farhan. At the end of the year, Rancho comes first in the class, while Farhan and Raju are last and second to last, respectively. During the yearly class photograph, Rancho makes a bet with Virus that if either Raju or Farhan gets a job from on-campus interviews, Virus will shave off his moustache.

Back in the present, Farhan, Raju and Chatur reach Shimla and are able to locate Rancho's house. However, they are shocked to come across another young man (Javed Jaffrey) identifying himself as "Ranchhoddas Shyamaldas Chanchad", whose face is pasted over Rancho's face in their graduation portrait with an ICE degree. When the confused Farhan and Raju ask Chatur how he got the address of this so-called "Ranchhoddas", he reveals that he got the address after his secretary came to Shimla to make a business deal with "Phunsukh Wangdu", a famous scientist and inventor who has 400 patents.

Upon learning this, Farhan and Raju confront the so-called "Ranchhoddas", who initially refuses to reveal anything and threatens them at gunpoint, but when they turn the tables and threaten to flush his late father's ashes down the toilet, he admits that he is the real "Ranchhoddas" and that the "Rancho" who was with them in ICE was actually their family's gardener's son nicknamed "Chhote", who was incredibly intelligent and gifted. After his parents' demise, Rancho was tasked by Ranchhoddas to complete all his academics.

When Ranchhoddas' father discovered this, he let the ruse continue for not wanting his son to experience the same disrespect that he experienced due to his illiteracy. Ranchhoddas went to London for four years while Rancho, who was interested in learning than in earning a degree, assumed his identity at ICE on condition that he cut off all contact with everyone there after graduation. However, Rancho warned Ranchhoddas afterwards that "two idiots" would come looking for him one day. Ranchhoddas tells Farhan and Raju that he is grateful for what all Rancho did and gives them his real address. Farhan, Raju and Chatur resume their journey to Ladakh where Rancho is apparently working as a schoolteacher.

The story settles back in their fourth year of college when the trio are approaching graduation. One night, Rancho points out to his friends why they consistently place last; Farhan's passion is wildlife photography, not engineering, and Raju lacks self-confidence. After Farhan and Raju make a bet that they will confront their problems if Rancho confesses his feelings for Pia, the trio drunkenly break into Virus' house at midnight. While Rancho climbs upstairs to propose to Pia, Farhan and Raju urinate on the letterbox of Virus, who spots Raju during the trio's escape.

The next day, Virus gives Raju an ultimate of either incriminating Rancho or being expelled himself. Not wanting to disappoint his family or betray his friend, a distraught Raju attempts suicide by jumping out from a 3rd-floor window and passes into a coma, but is successfully recuperated with the support of his family and friends over the next few months, while Virus also rescinds his rustication. With a newfound perspective of his life, Raju succeeds in securring a job with an engineering firm.

Meanwhile, Farhan also manages to convince his father to let him become a wildlife photographer. Thus, Virus' barber shaves off his moustache and Virus, in an attempt to protect his fragile ego, tries to sabotage Raju's chances of success by holding an unfairly different final exam. However, Pia drunkenly visits Rancho and hands him over the duplicate keys of Virus' office, encouraging him to copy the papers rigged by Virus for Raju.

The story catches up to the present where Farhan suddenly remembers Pia and learns that her wedding is taking place after he contacts at her home. Determined to take Pia along with them, Farhan and Raju gatecrash Pia's wedding with Suhas in Manali, Himachal Pradesh. Farhan sneaks in to reunite with Pia while Raju poses as a housekeeper to show that Suhas still has the materialistic attitude. Pia initially refuses to go with Farhan, despite hearing Suhas screaming at Raju for spilling mint chutney on his expensive wedding suit. Raju proceeds to fill in as the bridegroom and convinces Pia that she still loves Rancho. Pia eventually escapes with the duo and Chatur to go to Ladakh and reunite with Rancho.

As the story rewinds back to their final year of college, Rancho and Farhan break into Virus' office and successfully copy the question papers. Although Raju refuses to cheat, the trio are caught and expelled by Virus in the middle of the night. Pia then confronts Virus over her brother's suicide, which was prompted by similar pressure placed on him by Virus. That night, a heavy rainstorm cuts all the electricity and traffic and the now-pregnant Mona, who goes under labor pain, is unable to be driven to the hospital.

Applying their knowledge of engineering, the trio successfully conduct Mona's delivery by modifying a vacuum cleaner into a makeshift venthouse with the help of Pia through a webcam, along with the inverter that Rancho had created on the name of Virus beforehand. A grateful Virus acknowledges Rancho as an extraordinary student by gifting him a very rare Space Pen and revokes the trio's expulsions. On the graduation day, Rancho suddenly disappears and is never to be seen by his friends again.

As the story settles back in the present, Farhan, Raju, Pia and Chatur finally reach Ladakh where they are astonished to find a thriving school that is probably run by Rancho. They also run into Manmohan (nicknamed "Millimetre"), formerly an errand boy at the ICE, and now Rancho's assistant, who reveals to them that Rancho has keenly kept their careers up to date. Eventually, the four have a happy reunion with Rancho on a sandbar, where he admits that he is still in love with Pia and the two of them share a passionate kiss. Chatur assumes Rancho to be a mere schoolteacher and mocks him. However, when Pia asks Rancho his real name, he reveals himself, in a twisted ending, to be the successful scientist and inventor "Phunsukh Wangdu", whom Chatur's company is courting. A flabbergasted Chatur accepts defeat. The film ends with Rancho, Pia, Farhan and Raju running away laughing and a humiliated Chatur chasing after them.
