CEPI prioritises the World Health Organization's (WHO) "blueprint priority diseases", which include the Middle East respiratory syndrome-related coronavirus (MERS-CoV), Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), Nipah virus, Lassa fever virus, Rift Valley fever virus, Chikungunya virus, and the hypothetical unknown pathogen referred to as "Disease X". A fundamental requirement of CEPI's investment strategy is ensuring "equitable access" to vaccines during outbreaks, although recent policy changes may have affected this commitment. In 2022, CEPI set an ambitious vision to enable the rapid development of a new vaccine in response to pandemic threats within 100 days.
Conceived in 2015, CEPI was officially launched in 2017 at the World Economic Forum (WEF) in Davos, Switzerland. It was co-founded and initially funded with 46 crore US$ from the Bill and Melinda Gates Foundation, the Wellcome Trust, and the governments of India and Norway. Since its inception, CEPI has expanded its funding base, welcoming contributions from the European Union in 2019 and the United Kingdom in 2020. The organisation is headquartered in Oslo, Norway.

CEPI is a direct response to calls from four independent expert reports into the Ebola epidemic for a new system for stimulating the development of vaccines against epidemic threats. It was founded by the governments of India and Norway, the Bill & Melinda Gates Foundation, Wellcome and the World Economic Forum, which has played a key convening role, bringing together stakeholders at the 2016 Davos meeting and other events.
